Aquaporin-4 (AQP4) is the predominant brain water channel, shown to be involved in formation of edema after stroke. We have generated a transgenic mouse strain lacking AQP4 (AQP4-KO). In a mouse stroke model of middle cerebral artery occlusion, loss of AQP4 has shown to be protective. We have additionally generated a new mouse strain with a single mutation in the AQP4 gene. This mutation is described in a patient with signs of brain ischemia.

This project will focus on using a model for a smaller and more focal stroke (this procedure is approved in FOTS ID 14204, and has al lethality of around 5%) to investigate the effects of loss of AQP4, partial loss of AQP4 and the single mutation in the AQP4-gene. With this project we intent to develop new strategies in treatment of stroke.

To investigate the effect of loss of AQP4 we will need to perform experiments on 42 AQP4 KO, and for partial loss of AQP4 we will need 42 AQP4 heterozygotes. To study the effect of the partial mutation we will need 42 single mutation animals. For controls will need 84 wild-type animals. In total 220 animals.
